This article describes how to use a different DataDictionary file to add a custom enzyme to BioPharmaView™ software. To make this change, the BioPharmaView.Data dictionary file cannot be directly modified. This will result in a reinstall of the software. Locate the file ProteinPilot.DataDictionary.xml in the directory C:\Program Files\AB SCIEX\ProteinPilot and make a copy. If ProteinPilot™ software is not installed, then request a trial license from our technical support team.
- Open the ProteinPilot.DataDictionary.xml with Notepad or Notepad++.
- Create the digestion set by searching for "DIGEST_SET" in Notepad.
- Copy an existing digest set and insert it underneath the original digest set.
- Ensure the DIGEST_SET : XX (number is unique). Type in the enzyme.
- Search the document to ensure the three-letter code “TLC” for this enzyme is unique.
- Enter the amino acids and their unique cleavage attributes.
- The attribute table will tell you if you need to use CA or CB or CNB:
- Save this file and copy the ProteinPilot.DataDictionary file to C:\Program Files\AB SCIEX\BioPharmaView location.
- In addition, there will be a BioPharmaView.Data Dictionary file. This file cannot be modified. Please do not attempt to modify it otherwise the BPV software will need to be reinstalled.
- To confirm that the newly added enzyme can be viewed in BPV software, open any project and choose a peptide setting workflow.
